中文摘要: 动脉粥样硬化易损斑块的不稳定性是导致斑块内出血、破裂致动脉血栓形成从而引发卒中的主要发病机制。研发高效、灵敏、特异性好的分子探针,实现在体无创性早期识别动脉粥样硬化易损斑块,并进行早期干预,对预防心脑血管事件具有极其重要意义。本项目结合光声学成像技术优势及前期研究基础,拟构建靶向动脉粥样硬化易损斑块新生血管内皮生长因子受体(VEGFR)的光声分子探针,该分子探针携带光学吸收子-纳米金棒(gold nanorods, GNRs),可在近红外波段对动脉粥样硬化斑块内新生血管行光声分子显像,评估斑块稳定性;同时,该分子探针核心包载有相变材料(PFH),通过光致相变技术(ODV),可对不稳定性斑块内新生血管行血管损毁术,定向破坏易损斑块新生血管内皮细胞,使其成为多功能诊治分子探针,为动脉粥样硬化易损斑块在体、无创、早期诊断、治疗和疗效评价奠定了初步实验基础。

英文摘要: Atherosclerotic vulnerable plaque instability is the main pathogenesis of leading to plaque hemorrhage, causing rupture of the stroke induced by arterial thrombosis. Development of an efficient, sensitive and specific molecular probe to achieve in vivo and non-invasive, early recognition and early intervention of atherosclerotic vulnerable plaque is extremely important implications for the prevention of cardiovascular events. This study combines the advantages of photoacoustic imaging technology basis for preliminary studies to built a atherosclerotic vulnerable plaque neovascularization endothelial growth factor receptor (VEGFR) targeted photoacoustic molecular probe, such photoacoustic molecular probe was loaded with gold nanorods (GNRs), an optically absorbing materials which could be used for imaging the atherosclerotic plaque neovascularization in the near-infrared region to assess the stability of the plaque; Meanwhile, the core of the probe was package of phase-transition materials (PFH), using the optical droplet vaporization technology, the photoacoustic probe could be used to damage the neovascularization of the vulnerable plaque within the blood vessel, causing targeted disruption of vulnerable plaque neovascular endothelial cells. These functions make the novel theranostic molecular probe a promising tool for in vivo and non-invasive, for early diagnosis, treatment and evaluation of the efficacy of atherosclerotic vulnerable plaque, and this study provides the preliminary experiments foundation.
