中文摘要: 传染性法氏囊病(IBD)是主要危害雏鸡的急性、致死性、免疫抑制性传染病,免疫弱毒活疫苗是防控本病的主要手段,病毒滴度的高低一直是影响疫苗生产的关键因素。前期研究表明,IBDV弱毒疫苗株Gt 在DF1细胞上的复制滴度明显高于CEF细胞上的复制滴度,进一步研究表明eIF4AII在DF1细胞中呈低水平表达,而在CEF细胞中则高水平表达。为了探寻eIF4AII的差异表达是否是造成弱毒疫苗Gt在两种细胞上复制差异的原因,本项目以宿主蛋白eIF4AII和IBDV VP1为研究对象,利用蛋白质相互作用方法并结合蛋白上调、下调及RNA干扰等生物学技术,从IBDV感染CEF、DF1细胞后对eIF4AII的影响及eIF4AII对IBDV在CEF、DF1细胞上的复制及聚合酶活性影响等方面进行研究,揭示eIF4AII在IBDV的复制及其引起聚合酶活性差异中发挥的作用,从而也为IBDV疫苗生产提供理论指导。

英文摘要: Infectious bursal disease (IBD) is an acute, fatal and immunosuppressive disease primarily infecting young chickens. The use of attenuated vaccine is still the primary means to prevent and control this disease and the viral titers has been the key factor for the vaccine production. In our previous study, we found that the titers of attenuated vaccine strain Gt was significantly higher than that of CEF cells, and further studies shown that eIF4AII expressed lowly in DF1 cells, while had the high expression level in CEF cells. To explore whether the expression differences of eIF4AII causing Gt replicated differently in two kinds of cells, focalizing on the host protein eIF4AII and VP1 protein of IBDV, the project used methods of studying protein-protein interactions, overexpression, knockdown and RNA interference techniques to study the effects of IBDV to eIF4AII and eIF4AII to IBDV and polymerase activity, which leading to illustrate the effects to the replication and polymerase activity differences of IBDV. This study also provides theoretical guidance for IBDV vaccine production.
