中文摘要: 为了适应临床病原微生物检测需求,保障国家公共卫生安全,提高人民健康水平,建立能对病原微生物特征信息进行快速、多靶点检测且适用于自动化检测的分子诊断新技术十分必要,具有非常重要的实用价值和社会意义。本课题中,我们将综合使用多种功能化纳米颗粒,发展创新的靶序列制备技术和通用纳米颗粒芯片,突破现有检测方法的技术瓶颈,建立适用于多分子指标的高灵敏、高特异性、快速的病原微生物特征信息检测方法,并将之应用于实际样本检测。利用该方法与自动检测平台相结合能够实现病原微生物检测全过程自动化。在本课题研究中,我们强调技术的实用化,在方法学构建的同时兼顾建立标准化制备方法和质量控制体系,为此技术向临床转化打下坚实基础。本课题的实施将为我国感染性疾病的分子诊断提供具有国际竞争力的可靠工具,填补国内的相应空白。

英文摘要: To meet the needs of clinical detection of pathogenic microorganisms, ensure the safety of national public heath and promote the health level of people, it is very important to develop a new rapid, multiplex and automated pathogenic microorganisms detection platform. The development of this novel platform also has very important practical value and social significance. In this study, we will develop a novel pathogenic microorganisms detection platform with high-throughput, high-sensitivity, high-accuracy characteristics, and easy to operate on the commercial automated workstations. By using several kinds of functional magnetic nanoparticles and integrating innovative technologies on target preparation and beads array, we can break through the bottleneck of the existing pathogenic microorganisms biosensors. In this study, we also emphasize the practicability of the technology. We want the platform can be used in clinical application instead of stay on the paper. To make this method can be easily transfered to a clinical protocol, standardizing of preparations and establishing quality contrcol will be considered as the beginning of the methodology design. The implementation of this subject will provide a reliable and competitive tool for the development of molecular diagnostic of infectious diseases in our country and fill the blank of domestic.
