中文摘要: 副溶血性弧菌是水产品中重要食源性致病菌;代谢组学是通过定性、定量研究生物体内代谢物的变化来研究生物体代谢途径及生命活动特征的新兴交叉学科。目前从代谢组学水平深入揭示副溶血性弧菌致病机理及发掘其潜在生物标志物作为新的检测靶点的研究还鲜有报道。本课题拟开展致病性和非致病性副溶血性弧菌代谢物组比较分析研究,首先优化副溶血性弧菌代谢物组制备方法及分析方法,然后对致病性和非致病性副溶血性弧菌的挥发性和非挥发性代谢物组分进行电子鼻、GC-MS、LC-MS分析,生物信息学解析,比较其代谢物组异同,之后再深入研究致病性副溶血性弧菌在毒力基因(tdh或trh)处于不同表达量时的代谢物组异同。以获得副溶血性弧菌代谢轮廓,并期望找到其潜在生物标志物,为从代谢组学层面开发副溶血性弧菌新的检测靶点奠定坚实物质基础,为进一步阐明副溶血性弧菌致病机理奠定坚实理论基础。

英文摘要: Vibrio parahaemolyticus (VP) is an important foodborne pathogenic bacteria in seafood. Metabolomics is a functional genomics technology of vital importance for understanding cellular functioning because the metabolome is a direct reflection of the physiological status of a cell. There was few reports about the studies of the pathogenic mechanism and finding the specific biomarker of Vibrio parahaemolyticus based on metabolomics.Hence it is needed to study comparative metabolomics of pathogenic and non-pathogenic Vibrio parahaemolyticus. Firstly,a reliable cell quenching, metabolite extraction and analysis method will be optimized. Secondly, comparative metabolomics of pathogenic and non-pathogenic Vibrio parahaemolyticus will be deeply analyzed based on E-nose, GC-MS, LC-MS. Finally, metabolome of pathogenic Vibrio parahamolyticus at different expression level of virulence gene (tdh or trh) will be comparatively studied. The specific metabolite profiling of Vibrio parahaemolyticus may be summarized, and the potential biomarker of Vibrio parahaemolyticus may be found.This study will improve our understanding of the fundamental mechanisms of pathogenicity of Vibrio parahaemolyticus, and point to new potential diagnostic and therapeutic targets for Vibrio parahaemolyticus.
