中文摘要: 有关CpG纳米免疫复合制剂的开发和应用的研究在近年来取得了很大的进展。然而,纳米载体对于CpG寡核苷酸链与其靶受体间相互作用的影响依然很不清晰。本课题拟选取CpG-纳米金和CpG-DNA四面体两种纳米免疫复合制剂,对其与细胞界面的相互作用在多个层面,包括细胞膜、细胞器和关键蛋白等进行系统的研究;分析纳米免疫复合制剂的内吞途径和亚细胞分布,及其对于相关信号转导通路的调节作用;并考察纳米复合物的理化性质对于上述过程的影响,以期对于纳米免疫复合制剂的生物学效应和分子机理有更深入的了解。

英文摘要: Development and application of CpG-containing immunostimulatary nanoconjugates has been intensively studied. However, the influence of nano-carriers on the interaction between CpG oligonucleotide and its target receptor is unclear. In this project, we intend to explore the bio-nano interface between cells and two types of immunostimulatary nanoconjugates, CpG-AuNP and CpG-DNA tetra. We will systematically study the process of endocytosis and intracellular distribution of nanoconjugates, as well as their interaction with key proteins in signalling pathways. We will also investigate effects of their physicochemical properties on those interactions. These studies will provide new insight into the biological effects and cellular mechanisms of immunostimulatary nanoconjugates.
