中文摘要: 系统性红斑狼疮(SLE)是一种复杂的、确切发病机制不明的自身免疫性疾病。SLE有两个重要的分子特征:与粒细胞生成相关的转录因子和干扰素诱导基因的表达水平均显著增加。研究表明仅有I型干扰素( IFN)本身并不足以激发狼疮的全面发病,其他的致病因素也有可能参与SLE自身免疫的启动。我们最近发现一种特异的DNA-淀粉样纤维可以诱导中性粒细胞和I型干扰素(IFN)产生的pDCs,进一步诱导狼疮样疾病的发生。同时发现该狼疮样小鼠体内含有针对中性粒细胞特异蛋白的抗体。基于上述实验数据我们提出以下假设:中性粒细胞有可能是促进狼疮发病的免疫应答中的其中一环。因此,在本项目中我们将着重探索中性粒细胞如何和pDCs一起被DNA-淀粉纤维活化,其活化的确切模式是什么,同时明确中性粒细胞是否是导致该狼疮动物模型发病的不可或缺的关键因素。本项目研究结果将为我们理解启动自身免疫反应的关键环节提供重要线索。

英文摘要: The precise cause of systemic lupus erythematosus (SLE), a heterogeneous autoimmune disease with multiple organ involvement, is unclear. Two key molecular features of lupus are the upregulated transcription of genes involved in granulopoiesis and those induced by type I interferon. Recently, we found that a special type of nucleic acids-protein complex, DNA-containing amyloid fibrils, can attract neutrophils as well as interferon-producing pDCs, which corrects with the induction of lupus-like disease. Based on these observations, we formulate a novel hypothesis - neutrophils may be an integral part of the immune response that drives the development of lupus. In this proposal, we will test how neutrophils are activated together with pDCs by DNA-containing amyloid fibrils in any specific manner and check whether they are dispensable for the disease development in our animal model. This study will provide important clues to understand the key steps required to initiate autoimmune reactions.
