中文摘要: 瘤背石磺(Onchidium struma)是由海洋向陆地进化的标志性物种,其呼吸系统在整个生命周期中发生了很大的变迁,对其呼吸系统的研究可填补贝类研究的相关空白。本项目运用现代生物化学、生理学及分子生物学等手段,研究瘤背石磺的呼吸作用机制及血蓝蛋白基因结构与表达特征,采用组织切片和染液注射技术研究瘤背石磺呼吸腔的构造,并结合透射电镜观察其结构特征。用人工潮汐研究皮肤的呼吸效能,运用呼吸代谢测定装置研究瘤背石磺的呼吸速率,采用控氧密封系统研究瘤背石磺在不同氧浓度下的呼吸功能,并探索不同环境因子对瘤背石磺呼吸代谢的影响。通过分析瘤背石磺血蓝蛋白基因的分子进化特征,探索血蓝蛋白与瘤背石磺呼吸功能的对应关系,并与其他物种的进化地位进行比较,从而在理论上丰富瘤背石磺呼吸作用机制及其分子机理,完善瘤背石磺进化的理论体系。

英文摘要: Onchidium struma is a typical animal representing the transition from the ocean to the land. During its whole life cycle, its respiratory system has undergone great changes. So, it reflects the animal evolution from lower to higher.Study on the respiratory system of Onchidium struma will fill the research blank of Shellfish.In the present work, the structures of respiratory cavity of Onchidium struma are studied by tissue section technology, dye injection technology and transmission electron microscopy, and the skin breathing performance are analyzed by tidal experiments and the breathing rate are studied by respiratory metabolism measurement device. The respiratory function under different oxygen concentrations are studied by sealed control-oxygen system. Meanwhile, the effects of different environmental factors on Onchidium struma respiratory metabolism are also characterized. Furthermore, the molecular evolution of the hemocyanin gene is studied to explore the relations between hemocyanin and respiratory function and compare the evolutionary status of other species. Thus enrich the mechanism of respiration in Onchidium struma in theory and its molecular mechanism, perfect theoretical system of Onchidium struma evolution.
