中文摘要: 2006年由高致病性猪繁殖与呼吸综合征病毒变异株(HP-PRRSV)引起的"猪高热病"首先在我国南方部分地区暴发,并迅速在全国蔓延,此后,该病持续流行,给我国养猪业造成了极大灾难。研究表明HP-PRRSV与细菌的协同作用是导致"高热病"高死亡率的重要原因之一。然而HP-PRRSV与细菌协同致病作用的分子机制目前尚不清楚。本研究借鉴国外学者已经建立的PRRSV-LPS感染模型,通过研究 HP-PRRSV感染对LPS/TLR4活化的正向调控机制,鉴定HP-PRRSV基因组中发挥协同致病作用基因靶点,阐明HP-PRRSV与细菌协同作用机理,解析HP-PRRSV引起机体高热及高死亡率的分子机制,为临床防控该病提供理论依据。

英文摘要: Since 2006 there have been outbreaks of a severe form of PRRS in South China characterized by prolonged high fever. It has rapidly become endemic all of the China and causes devastating economic loss from then on. Several studies have confirmed that the causative agent of these outbreaks was a highly pathogenic strain of PRRSV (HP-PRRSV), and that secondary bacterial infection may also contribute to the high mortality. However, the molecular pathogenesis by which the HP-PRRSV and bacterial action in synergy is unknown. In this study, to determine the mechanisms by which HP-PRRSV and bacteria synergistically act for the occurrence of the characteristically high fever and high mortality, we used an in vitro model according to the previous report of foreigner scholars to investigate the positive regulation of LPS/TLR4 activation. The results will provide a prerequisite theoretical basis for better controlling and therapy of the disease.
