中文摘要: 为了探讨壳聚糖对断奶仔猪氧化应激反应的缓解作用及其调控机理,本项目利用体内法研究壳聚糖对氧化应激状态下断奶仔猪生长性能、免疫功能、抗氧化功能及肝脏、脾脏、小肠组织病理变化的影响,并借助分子生物学手段检测壳聚糖对氧化应激状态下断奶仔猪肝脏、脾脏和肠道组织中核转录因子NF-κB、转录激活蛋白AP-1(包括c-Fos蛋白和c-Jun蛋白)以及超氧化物歧化酶SOD、谷胱甘肽过氧化物酶GSH-Px、过氧化氢酶CAT的基因表达的影响;同时利用体外法研究建立断奶仔猪淋巴细胞体外氧化应激模型,并采用该模型研究壳聚糖对断奶仔猪淋巴细胞氧化应激的抵抗作用,同时利用抑制剂阻断法从NF-kB和AP-1途径探讨壳聚糖对断奶仔猪氧化应激缓解作用的分子调控机理。项目的完成对于深入了解壳聚糖对断奶仔猪氧化应激的缓解作用及其调控机理具有重要的学术价值,在断奶仔猪生产中具有重要的实践指导意义。

英文摘要: In order to explore the mitigation action of chitosan on oxidative stress of weaned piglets and the underlying mechanism, in vivo experiments are designed to study the effects of dietary chitosan on growth performance, immune function, antioxidative function and pathological change of liver, spleen and small intestine in weaned piglets under oxidative stress, and detect the gene expression of NF-κB, AP-1 (including c-Fos and c-Jun), SOD, GSH-Px and CAT in these tissues. Meanwhile, in vitro experiments are designed to establish the optimal model of oxidative stress of lymphocytes in piglets, and based on the model to study the resistance action of chitosan on oxidative stress in lymphocytes, in addition, to erplore the molecular mechanism of mitigation action of chitosan on oxidative stress of weaned piglets via NF-κB pathway and AP-1 pathway, respectively. The results of the project would have a high academic value for revealing the mechanism of the mitigation action of chitosan on oxidative stress of weaned piglets, and show a great significance in the production of weaned piglets.
