中文摘要: 针对每年国内外果品采后损失严重的实际问题,开展果实成熟衰老机制研究具有重要的理论和实际意义。番茄是重要的园艺作物和研究果实成熟的模式植物,课题组前期研究表明small RNA合成中重要核酸酶SlDCL2c的表达与番茄果实成熟相关。本项目拟对SlDCL2c开展深入研究,通过转基因、RNA-seq和Small RNA-seq等技术,解析SlDCL2c在番茄果实成熟中的生物功能及作用途径;利用瞬时表达、Co-IP和体外重建等手段,在生化水平上探明SlDCL2c识别和加工果实成熟相关small RNA前体的生物学规律;最终准确而清晰的诠释SlDCL2c调控果实成熟的分子机制,为果实发育成熟调控提供新的基因资源,为改造番茄果实性状提供新的途径。

英文摘要: Due to serious loss of fruits at home and abroad each year, it is important theoretical and practical significance to launch the research on the mechanism of fruit ripening and senescence. Tomato is an important horticultural crops and a model plant for the research in fleshy fruit ripening, our previous study shows that SlDCL2c expression is related to tomato fruit ripening, which is responsible for the biosynthesis of small RNA. This project aims to carry out in-depth research on SlDCL2c, and to illustrate biological function and regulation pathway of SlDCL2c in tomato fruit ripening by the methods of transgenic, RNA-seq, Small RNA-seq and so on; to verify biological rule of identification and processing of small RNA precursors by SlDCL2c at the biochemical level via the technologies of transient expression, Co-IP, in vitro reconstruction and so on; finally to clarify the molecular mechanism of SlDCL2c regulation on fruit ripening, which will provide new gene resource and new way for fruit ripening regulation.
