中文摘要: 番茄是世界及中国最主要的蔬菜之一,番茄晚疫病给番茄生产造成严重危害。番茄晚疫病抗性育种一直是国内外研究的重点和热点。本研究以高抗番茄晚疫病自交系'CLN2037E'和感病自交系'5#自交系'为试材,运用高抗番茄晚疫病诱导cDNA文库、番茄基因组CDS数据库、晚疫病抗性相关Unigene、晚疫病诱导转录组数据,并结合生物信息学方法,发掘晚疫病抗性基因。之后运用实时荧光定量PCR技术,鉴别响应晚疫病的基因。最后通过基因过表达途径转化感病番茄自交系和VIGS技术转化抗病自交系,对获得的阳性植株进行晚疫病接种试验,评估抗病能力的变化,鉴定基因的功能。研究结果将初步阐明抗病基因与番茄晚疫病抗性之间的相关性,为进一步探明番茄晚疫病的抗性机制及晚疫病抗病育种提供理论依据和基因资源。

英文摘要: Tomato is one of the most important vegetables in the world. Tomato late blight (LB) caused seriou yield and quality losses in tomato prodution. Resistant breeding against LB has been of interest for many years. In this study, highly resistant inbred line CLN2037E and susceptible inbred line 5#were used as experimental materials. Based on the data of differential cDNA library, late blight resistance-related unigene and tomato CDS database, transcriptome data induced by late blight, late blight resistance genes were excavated through the bioinformatics technology. Next, real-time PCR was used to identify the resistance genes from the excavated genes. At last, ectopic expression and RNA interference were employed to transform the line 5# and CLN2037E, respectively. Positive plants were inoculated by late blight fungus to evaluate the genes' function. The results will preliminarily illustrate the correlation between the resistance to late blight and the resistant genes, and provide the theory basis and gene resource for the resistance mechanism and resistance breeding, respectively.
