中文摘要: 促进白色脂肪向棕色脂肪转分化已成为肥胖治疗的新途径。uc.342是我们应用芯片技术筛选白色脂肪与棕色脂肪组织间差异lncRNA时发现的、差异高表达于棕色脂肪组织、物种间高度保守、功能未知的一条lncRNA。已发现:uc.342在白色脂肪前体细胞中过表达可促进白色脂肪向棕色脂肪转分化,软件预测及实验发现HOXC8为uc.342的潜在靶标,而文献报道HOXC8在白色脂肪向棕色脂肪转分化中起重要作用,提示uc.342可能通过调节HOXC8促进白色脂肪向棕色脂肪转分化。研究拟进一步阐明uc.342的生物学特征、在白色脂肪向棕色脂肪转分化中的具体功能;以HOXC8为机制线索,论证uc.342影响白色脂肪向棕色脂肪转分化的机制;并在体评估uc.342用于肥胖治疗的可行性。迄今未见uc.342参与白色脂肪向棕色脂肪转分化的报道,研究将为白色脂肪向棕色脂肪转分化提供新机制,为肥胖治疗可能提供新靶标。

英文摘要: Turning white adipocyte into brown adipocyte becomes hope for obesity treatment. LncRNA uc.342 is a highly reserved BAT specific lncRNA identified from differential lncRNA expression profile between mouse white adipocyte tissue and brown adipocyte tissue. Our further studies revealed overexpression of uc.342 in white preadipocyte could induce white-to-brown adipocyte transdifferentiation.Bioinformatic analysis and experiments suggested HOXC8 was candidate target gene to uc.342. Based on these data,we plan to investigate the role and function of uc.342 in white-to-brown adipocyte transdifferentiation by analyzing morphology,biological characteristics,and detailed regulatory mechanism of the interaction between uc.342 and HOXC8.Furthermore,we will evaluate the effect of uc.342 in treating obesity mouse. So far there is no report about lncRNA in adipocytes' transdifferentiation. This study will show us a new target and method to combat human obesity.
