中文摘要: TRAIL 能特异性地诱导大部分肿瘤细胞凋亡,但部分肿瘤细胞对它耐受。我们以前的研究结果表明:抗凋亡蛋白c-FLIP(L)的持续表达是前列腺癌细胞耐受TRAIL的必要和充分条件;并首次阐明c-Fos迅速转入细胞核内是肿瘤细胞对TRAIL敏感的必要条件,c-Fos及其转录复合物AP-1可通过结合在c-FLIP(L)启动子的特定部位而拟制c-FLIP(L)的表达,从而使肿瘤细胞对TRAIL敏感;首次发现c-Fos/AP-1的诱导剂TPA和稳定剂MG-132通过激活c-Fos/AP-1逆转前列腺癌细胞的TRAIL耐受。这些结果肯定了c-Fos/AP-1- - c-FLIP(L)这一新通路在调控肿瘤细胞凋亡中的作用。而这一通路中最关键的一个分子事件是:c-Fos迅速进入细胞核并发挥作用。本课题的目标就是研究肿瘤细胞中c-Fos在TRAIL刺激下进入细胞核的机制,期望更深入地了解肿瘤细胞的凋亡机制。

英文摘要: TRAIL can induce apoptosis in most cancer cells but sparing normal cells. However, some cancer cells are resistant to TRAIL. Our previous studies have demonstrated that: a) the persistent expression of c-FLIP(L) is necessary and sufficient for prostate cancer cells to be resistant to TRAIL; b) for the first time, c-Fos rapid translocation into nuclear is required for prostate cancer cells to be sensitive to TRAIL. Furthermore, c-Fos/AP-1 promotes TRAIL-induced apoptosis via binding to the specific site of c-FLIP(L) promoter and inhibiting the expression of anti-apoptotic c-FLIP(L); c) c-Fos/AP-1 inducer TPA and MG-132 can reverse TRAIL resistance via activating c-Fos/AP-1. These results confirm the role of a novel pathway c-Fos/AP-1- - - - c-FLIP(L) in regulating apoptosis of tumor cells. The most critical event in this novel pathway is how c-Fos enters nuclear and plays its apoptotic role. The aim of this proposal is to explore the molecular mechanisms of translocation of c-Fos under TRAIL stress.
