中文摘要: 新生隐球菌感染发病率逐渐增加,其诊断和治疗均存在巨大挑战。深入研究抗新生隐球菌感染的免疫应答机制具有重要的生物学意义和医学应用价值。研究表明,Th1和Th17型应答在清除隐球菌中起关键作用。最近,能够分泌IL-9的Th9亚群的发现使IL-9成为许多研究领域的热点。Th17据报道也可产生IL-9。IL-9可以作用于多种免疫细胞,其功能复杂多样。迄今,有关IL-9在抗隐球菌免疫应答中的作用尚无任何报道。我们的预实验结果发现隐球菌株感染动物肺脏后可以显著诱导IL-9的表达,但其是由Th9还是Th17分泌尚不清楚。我们的结果还提示IL-9可能起到保护性的作用,但其作用和机制有待深入研究。因此,本研究拟采用不同毒力隐球菌株诱导不同免疫应答动物模型及细胞模型,在体内和体外对IL-9在抗隐球菌免疫应答中的作用和机制进行深入研究。本研究旨在丰富机体抗隐球菌免疫应答的理论,并为免疫干预隐球菌感染提供新思路。

英文摘要: The incidence of cryptococcosis is gradually increasing, and its diagnosis and treatment are now still great challenge. Further investigation on the mechanisms of host immune response against Cryptococcus neoformans infection has very important biological significance and practical value of medicine. There have been studies suggested that Th1 and Th17 responses play a key role in the clearance of Cryptococcus neoformans. Recently, since the discovery of IL-9-producing Th9 helper cells, IL-9 has become a very active hot spot in many scientific research fields. It is reported that Th17 cells can also produce IL-9. IL-9 acts on various immune cells and exhibits pleiotropic effects. To date, no reports have been published concerning the role of IL-9 in the control of immune response against Cryptococcus neoformans. Our preliminary studies have showed that Cryptococcus neoformans infection can strongly induce the expression of IL-9 in lungs, but little is known about which cells (Th9 or Th17?) are responsible for IL-9 secretion. Our results also suggested that IL-9 may play a protective role, whereas the effect and mechanism of IL-9 in immune response against Cryptococcus neoformans need further investigation. Therefore, this study aims to investigate the effect and mechanism of IL-9 in immune response against Cryptococcus neoformans both in vitro and in vivo by using the immune response models induced by different virulent Cryptococcus neoformans strains and cell line models. This study will provide new fundamental theory for host immune response against Cryptococcus neoformans, and supply new ideas for immunological intervention of Cryptococcus neoformans infection.
