中文摘要: 骨关节炎(Osteoarthritis, OA)是一种退行性关节病变,其主要病变以关节软骨的退行性变为主要特征,可以造成巨大的社会和经济负担,但发病机制仍然不清楚。国内外研究结果发现TGFβ和RhoA/ROCK信号通路在OA发病中发挥着重要作用,但是目前关于两者在OA病变过程中的相互关系及调控机制研究较少。本研究以SD大鼠为研究对象,应用外源刺激因子诱导信号通路激活、抑制、RT-PCR、western blot、FRET技术等分子生物技术相结合,通过体内、体外两部分实验,多角度证明TGFβ与RhoA/ROCK信号通路在OA病理变化中的相互作用关系,旨在明确TGFβ对RhoA/ROCK信号通路调控的分子机制,进而为研究TGFβ对RhoA/ROCK信号通路调控治疗OA病变提供线索和依据,为开发OA治疗干预药物提供实验依据。

英文摘要: The key pathophysiological features of osteoarthritis(OA) joints include gradually degeneration of the articular cartilage.incidence of OA cause a large social and economic,howerer it is still no clear about the pathogenesy of OA. Recent stydy results show TGFβ and RhoA/ROCK signal pathway play the important role in etiology of OA. Howerer,mechanisms about TGFβ and RhoA/ROCK pathway was studied less. In this study,using SD rats as object of study, We will reveal the relationship between interact of TGFβand RhoA/ROCK pathway in the process of pathological change about OA in vivo and in vitro, some molecular biological technology are utilized in this study, such as stimulation of activating agent and inhibitor about TGFβand hoA/ROCK pathway,real-time RT-PCR, western blot, fluorescence resonance energy transfer. The results of this study will reveal the interact relationship of TGFβand RhoA/ROCK pathway which mediate generation and degeneration of the articular cartilage to affect pathogenesy of OA ,to elucidate the regulation mechanism between TGFβand RhoA/ROCK pathway. Our study may bring clues to illuinnate the pathogenesis of osteaarthritis diseases,and these may provide new perspective for therapeutic strategies fof osteaarthritis.
