中文摘要: 神经根性疼痛是神经根型颈椎病(CSR)的主要症状,《黄帝内经》论述:气伤痛,尽气闭环,痛病必下,基于气行不畅的气伤痛理论较基于经脉闭塞的不通则痛理论更契合痛症病机。JAK-STAT/SOCS信号通路介导了CSR疼痛机制,电针治疗CSR目前以夹脊穴局部取穴为主,未能体现针灸疗法远端取穴的优势。基于气伤痛理论,从该信号通路交互作用角度探讨远端腧穴电针镇痛机制的研究缺乏。项目将构建CSR大鼠模型,与正常组、调气电针组对照,通过痛阈检测,采用ELISA法、RT-PCR、Western Blot等方法观察大鼠脊髓及神经根组织中JAK-STAT/SOCS信号通路关键蛋白及下游因子IL-1β、IL-6、PGE2、COX-2等表达的变化,探讨基于气伤痛理论远端取穴的调气电针对CSR大鼠模型JAK-STAT/SOCS信号通路交互作用的影响,揭示其作用靶点及镇痛机制。

英文摘要: Radicular pain is the main symptom of cervical spondylosis radiculopathy (CSR), Huang Di Nei Jing states: Qi shang tong ,Jing qi bi huang,tong bing bi xia.The theory of Qi shang tong based on Qi running sluggish is more in line with the pain pathogenesis than the theory of Bu tong ze tong based on the meridians occlusion. Study confirmed JAK-STAT/SOCS signaling pathway mediated by CSR pain mechanisms, Electroacupuncture(EA) treatment CSR currently dominated Jiaji local points which is can not reflect the distal acupoints acupuncture advantage. Based on the Qi shang tong theory to explore the distal acupoints EA analgesia mechanisms from the interaction of the signal path is lacking. The project will build a rat model of CSR, compare to the normal group, EA group. Use the pain threshold testing and ELISA, RT-PCR, Western Blot and other methods, observe the expression of JAK-STAT/SOCS signaling pathway key protein and IL-1β、IL-6、PGE2、COX-2 in rat spinal cord and nerve roots organization. The purpose of this study is to discuss the influence to CSR rats with distal acupoints EA base on qi shang tong theory by studying the JAK-STAT/SOCS signaling pathway interactions, and reveal its targets and analgesic mechanisms.
