中文摘要: 新技术和新方法的发展一直是推动蛋白组学研究的关键和强大动力,研制高效低残留的新型蛋白质样品处理和分离材料,对蛋白组学研究意义重大。本项目拟将点击化学反应应用在蛋白质样品处理和分离材料制备中。通过点击化学反应对磁性纳米材料和整体柱材料进行表面修饰,获得高特异选择性的目标蛋白质分离富集新材料新方法。制备表面含有点击反应活性基团的磁性材料和整体柱骨架,再点击反应制备核酸适配体、蛋白质分子印迹聚合物、硼酸修饰的磁性纳米材料和核酸适配体、硼酸共价色谱整体柱材料,并应用于复杂生物样品(血样、尿样、组织样品)中目标蛋白质的分离富集。研究点击化学在磁性纳米和整体柱材料表面定向修饰特异性配基的通用性及反应条件,优化蛋白质分离条件。本项目的研究将丰富点击化学、材料的表面修饰和核酸适配体的内容,有望获得具有自主知识产权的蛋白质组样品处理和分离材料制备新方法、新技术,具有重要的理论研究和实际应用价值。

英文摘要: The development of novel techniques and methods is the strong motivation to promote the research on the proteomics. It is of significance to prepare novel materials for treatment and isolation of proteins. In this project, the methods of the preparation of novel magnetic and monolithic column materials for the high specific isolation and enrichment of target proteins via click chemistry will be developed. Firstly, the 'clickable' active groups are modified on the surface of the magnetic nanomaterials and framework of monoliths; subsequently, magnetic nanomaterials functionalized by the specfic liands such as aptamer, molecularly imprinted polymers, boronate and polymer monoliths functionalized by aptamer and boronate are obtained via click chemistry. The resultant magnetic and monolithic column materials are applied to isolation and enrichment of proteins from the biological samples (e.g. blood, urine, tissue) with complex matrix. In this project, the reaction condition of immbolization of aptamer, molecularly imprinted polymers, boronate on the surface of magnetic and monolithic column materials will be studied in detail.
