中文摘要: 缺乏突破性育种材料和功能基因是导致目前我国小麦生产发展缓慢、单产增幅不高的根本原因和主要障碍。本项目从产量三要素影响因子千粒重入手,对实验室前期获得的产量相关候选基因TaBS3和TaWox2的功能进行深入分析研究,通过转基因小麦植株、全基因组关联分析以及候选基因组织时空表达特性分析等,明确候选基因在籽粒发育过程中的调控特性,其与粒重以及小麦产量的关系;进一步采用酵母双杂和高通量转录组测序技术,大规模挖掘候选基因的互作蛋白以及下游调控基因蛋白(拟以转基因过表达植株和对照植株为试材)。通过功能验证和互作研究,有望解析TaBS3或TaWox2调控籽粒发育的可能机理、初步明确与候选基因互作的籽粒发育调控网络因子,为小麦单产提高和分子育种提供可靠的基因资源和小麦材料。研究成果具有十分重要的科学意义和实际应用价值。

英文摘要: Lacking of the critical breeding parents or function genes is one of the reasons and main obstacle to the development of wheat production in China. This project aims to the function and regulation mechanism analysis of candidate genes (TaB3 and TaWox2) related to grain yield on 5DS in common wheat. First, we combine the transgenic technique, genome-wide association analysis as well as the expression patterns of candidate genes in different wheat tissues and at the different stages of kernel development (7 days,14 days, 28days after flowering) together to know exactly the role of candidate in wheat kernel development. Then, select and identify the interaction proteins and downstream regulatory genes of the candidate genes by the method of Yeast-two-hybrid and RNA-seq respectively. The function of differentially expressed downstream genes will be analyzed by bioinformatics and other molecular biology techniques. Last, transgenic research will be performed to verify their functions of downstream genes in the grain development. According to these results, the candidate genes working model linked with wheat grain development will be constructed. This project will contribute to clarify regulatory network of wheat grain development, and provide new gene elements for the molecular breeding of wheat.
