中文摘要: 构建高灵敏和高选择性的核酸传感平台在临床诊断、遗传分析、环境监测和生物反恐等领域具有重要意义。本项目首次提出将DNA水凝胶和电纺纳米纤维整合制备一种新型DNA水凝胶-电纺纳米纤维复合新材料,研究DNA水凝胶-电纺纳米纤维复合材料作为新型传感材料的传感机制及规律,据此发展和建立面向肿瘤和遗传疾病相关核酸序列的高灵敏、高选择性的分析检测新方法,并拓展新方法在疾病诊断和遗传分析等领域中的应用。本项目充分结合了DNA水凝胶易于功能化、可操控性强与电纺纳米纤维易于制备、高比表面积等优势,将拓展DNA水凝胶材料和电纺纳米纤维材料在生物传感领域中的应用,并有望实现实际样品中肿瘤与遗传疾病相关特定核酸序列的分析检测。

英文摘要: The sensitive and selective detection of nucleic acids is important in clinical diagnosis, genetic analysis, environmental monitoring, and biological antiterrorism. In this project, we will integrate for the first time the DNA hydrogel and the electrospun nanofibers to fabricate the DNA hydrogel-electrospun nanofiber composite material. Based on this new composite material, we will develop a nucleic acid sensing platform with high sensitivity and selectivity for tumors and hereditary diseases-related specific nucleotide sequences. The constructed sensing platform for nucleic acids combines the advantages of easy functionalization and good designability of DNA hydrogel and the large surface area-to-volume ratio of electrospun nanofibers. This project will provide a new route for the application of DNA hydrogel and electrospun nanofibers in biosensing, and a promising strategy for highly sensitive detection of specific nucleotide sequences in biological samples.
