中文摘要: 高效抗逆转录病毒治疗(HAART)可抑制病毒的复制,将血浆病毒载量降低至现有常规检测方法测不出的水平,且使HIV感染者的病死率明显降低。然而HAART很难根除HIV病毒,这是由于病毒潜伏库的存在。本项目将根据HIV潜伏性感染的分子生物学机制,分别建立基于T细胞增殖和细胞毒性T淋巴细胞(CTL)免疫应答的潜伏感染模型,理论上分析模型的动力学性态;利用医学文献中已有数据,运用敏感性分析方法找到影响病毒反弹的关键参数,并进行参数估计;分析不同水平的药物疗效和T细胞增殖项对病毒潜伏库的持续存在和病毒反弹的影响;探讨CTL细胞免疫应答清除病毒潜伏库的可能性,并给出影响病毒清除的因素。

英文摘要: Highly active antiretroviral therapy (HAART) can suppress HIV replication, reduce plasma viral load in HIV patients to below the detection limit of conventional clinical assays and decrease the morbidity, but can not eliminate the virus because HIV establishes latent reservoir. In this project, based on the mechanism of HIV latent infection, we will formulate HIV latent infection models with T-cells proliferation and CTL immune response respectively, and will analyze the dynamical behavior of these models theoretically. Using the estimated data of the medical literatures and sensitivity analysis method, we are trying to find the key parameters which impact viral blips, and estimate these parameter values. Then, we discuss the effects on HIV latent reservoir persistence and viral blips with different levels of drug efficacies and T-cells proliferation. Furthermore, the possibility of CTL immune response to clear viral latent reservoir is explored and the factors on virus eradication are studied.
