中文摘要: 高密度脂蛋白(HDL)是公认的动脉粥样硬化(As)防御因素,临床上常规检测HDL-胆固醇(HDL-C)水平作为As风险性的评价指标,但HDL-C反映的HDL代谢池而不能完全准确地评价HDL抗As的功能。针对HDL特异性结构蛋白及相关功能的研究将有利于新的As检测靶标的发掘及应用。我们前期针对As发生发展中HDL "失功能性"的研究发现:HDL专有抗氧化酶- - 对氧磷酯酶1(PON1)的活性与HDL抗氧化抗炎的功能状态密切相关,在转基因动物As模型上PON1的活性和HDL的炎症状态可直接对As的发生发展进程进行评估。考虑目前尚未建立简便稳定、统一标准的检测方法,且人群中存在个体差异等因素,本课题拟改良优化已建立的"PON1功能活性"及"HDL抗炎指数"的检测方法,并在冠心病等As性疾病的群体中进行相关性评价,从而揭示新的针对HDL功能的检测靶标用于临床As诊断、风险评估和临床治疗监测可行性。

英文摘要: Plasma High-density lipoprotein is well established as a protective factor for the development of atherosclerosis. Although the level of HDL-cholesterol (HDL-C) as a routine test in clinical can relect the risk of As, HDL-C reflects the HDL metabolic pool that can not be entirely accurate evaluation of the function of HDL。Focus on HDL-specific structural proteins and related functions will be conducive to the new As detection targets discovery and application. In our previous studies of structure and function of "dysfunctional" HDL in As model animals, we found that anti-inflammatory antioxidant capacity of dysfunctional HDL decreased, and the risk of As and the development of state could be reflected by HDL anti-inflammatory index and the functional activity of HDL proprietary antioxidant enzymes - paraoxonase 1 (PON1). This project intends to combine the individual differences and other relevant factors, and test methods for the establishment and optimization of HDL function in the target groups for Coronary heart disease, etc. As disease ,which will reveal "functional activity of PON1" and "HDL anti-inflammatory index" as new detection targets for clinical As diagnosis, risk assessment and feasibility of the clinical monitoring of therapy.
