中文摘要: 近年来利用多亲本衍生群体开展遗传研究的重要性日益突出,但多亲本群体中遗传连锁分析和基因定位方法还不完善。作物育种中涉及四个亲本时,双交设计是一种常用的交配方式。本项目将对双交F1及其衍生加倍单倍体和重组近交家系群体的遗传分析方法进行系统研究,具体内容包括重组率估计、连锁图谱构建与整合以及基因定位方法,利用模拟群体验证这些方法的有效性,在软件QTL IciMapping中实现双交F1及其衍生群体连锁图谱构建和基因定位功能,并将这些方法应用于二至三个真实双交F1或其衍生群体。本项目研究结果将为更多亲本衍生遗传群体的分析方法提供借鉴;在无性系繁殖的物种中,两个品种间杂交产生的F1群体可以看作一个双交F1群体,因此本研究的结果还可为无性系繁殖物种(如马铃薯、甘薯、以及大量的林木和花卉等)的遗传研究提供有效方法和工具。

英文摘要: Genetic populations derived from multiple parents have been proposed and become more and more important now. But the corresponding methods of linkage analysis and gene detection have not been well studied. When there are four parents, double cross design is commonly used in plant genetics and breeding. In this project, we will focus on the genetic analysis methods in double cross F1 population, and derived doubled haploid (DH) and recombination inbred lines (RIL). Our objectives are (1) to derive the maximum likelihood estimation of recombination frequency; (2) to develop an efficient algorithm for linkage map construction; (3) to develop the genetic and statistic analysis method for gene detection of quantitative traits. Simulation study will be conducted to demonstrate the efficiency of these methods. Functionalities for map construction and gene detection in double cross F1 and derived populations will be implemented in the software QTL IciMapping. These methods will be applied in two or three actual double cross F1 and derived populations. Outcomes from this project will provide useful information for the genetic analysis in populations derived from more parents.
