中文摘要: 莲(Nelumbo nucifera)为睡莲科的著名水生花卉,栽培历史悠久,种质资源丰富,重要的观赏性状包括植株大小、花色、花型均有丰富的变异,是研究观赏性状分子遗传规律的理想材料,本课题将围绕株型这一数量性状进行研究。课题组前期用大株重瓣莲品种'宝珠观音'及小株少瓣莲品种'恋夏'作为杂交亲本,构建了F1代拟测交群体,本项目在此基础上通过不同的分子标记分析,构建连锁遗传图谱,并初步定位株型QTL。然后进一步建立"拟F2"代作图群体以及运用BSA法进行精密作图和QTL精细定位。对收集的400个莲种质群体,结合表型与分子标记研究进行株型性状的关联分析。然后将株型QTL定位和关联分析结果进行比较验证,高效精确的获取与株型相关的QTL或候选基因,最后对其开展基因功能互补试验和时空表达分析,明确候选基因的功能与调控方式,初步探寻莲株型调控的分子机制,为最终进行莲花及其它花卉植物株型改良奠定基础。

英文摘要: The Nelumbo nucifera is a famous aquatic flower with a very long history of cultivation in China. The germplasm resources are abundant including many cultivars with different variations of important ornamental traits-such as plant size, flower color and flower shape. Thus it is a perfect material for studying on molecular genetic mechanism of ornamental traits. The quantitative trait-plant size of lotus will be studied in this project. We have previously obtained "two-way pseudo-testcross" F1 population by crossing between a large plant size of cultivar- - 'Baozhu Guanyin' and a small plant size of cultivar- - 'Lian Xia'. In the present project, using different molecular markers to detect polymorphism of F1 population the genetic linkage map can be constructed and then the QTLs encoding plant size will be preliminarily located in the linkage map. Furthermore through construction of the "two-way pseudo-testcross" F2 population and application of BSA, a precise linkage map will be constructed with the precise location of QTLs. Besides, association mapping will be carried out on 400 lotus germplasms with molecular markers analysis as well as phenotypes measuring. Based on the comparison of linkage mapping and association analysis, the QTLs or candidate genes related with the plant size will be identified accurate
