中文摘要: 膜蛋白受体是人体内关键的一类蛋白质,对于药物开发有着重要意义。然而膜蛋白受体结构的复杂性导致其成为了信息科学技术在其应用的主要瓶颈,传统计算方法目前已无法满足其要求。本项目面向膜蛋白受体,建立一套完整的蛋白质结构与网络预测与分析系统;基于新颖的协同进化算法,改进了传统的机器学习和智能计算方法,建立膜蛋白受体三级结构的预测模型;基于协同进化的多参数优化,为膜蛋白受体的相互作用与网络的构建,调控和优化提供技术手段;筛选出核心的膜蛋白受体,探索其传导信号的过程,探讨生物实体网络的普适性规律;对各种智能算法在膜蛋白受体及其网络的表现进行评估,并开发一套完整的面向用户的在线预测分析系统。综上所述,本项目将研究成果将对进化计算与生物信息学交叉学科的发展与药物研发技术作出贡献,同时为新型智能算法在复杂蛋白质上的应用奠定理论与技术基础。

英文摘要: Membrane protein receptors are key proteins in human genome, which are significant for drug discovery. However, the structural complexity of the membrane protein receptors makes it a major bottleneck of the application of informatics. The present project mainly focuses on membrane protein receptors, develop a prediction and analysis system. Based on the novel co-evolutionary computation, the project improves traditional machine learning and intelligent computation methods, constructs a computational model for the protein tertiary structure prediction of the membrane protein receptors; analyzes the controllability and optimization of the network for reconstructing membrane protein receptor interaction network; selects the Hub protein, explores the pathway of signal transaction and the general rules of biological networks; evaluates the performance of evolutionary computation and other intelligent algorithms and develops a Web server of membrane protein receptor prediction system. Generally speaking, the results of the present project will contribute on the bioinformatics and drug discovery industries, and build the theoretical and technical evidence for the novel intelligent algorithm, which is used for studying the proteins with complex structure.
