中文摘要: 明确胃癌细胞转移和侵袭机制,对于临床治疗和预后判断具有重要意义。研究发现,抑癌基因DNA甲基化等表观遗传学改变可导致基因表达沉默,由此赋予癌细胞更高侵袭性或转移优势生长。我们前期应用基因芯片对胃癌前哨淋巴结转移前后表达谱进行分析,发现多个基因转移后表达沉默。通过亚硫酸氢盐修饰PCR 测序分析,我们首次发现表达沉默的NPR1 基因启动子区存在CpG 岛并在胃癌细胞中存在甲基化畸变。本项目即在此研究基础上,采用启动子荧光素酶报告分析、去甲基化药物处理和免疫沉降的方法,分析NPR1 基因表达沉默的表观遗传学机制;同时构建NPR1 全长mRNA 表达载体并转染到表达沉默胃癌细胞中,对细胞进行克隆成形分析、侵袭分析和转移分析,确认NPR1 基因在胃癌转移中的关键作用;最后大样本分析NPR1 基因甲基化畸变和临床进展和预后的关系,从而系统阐明NPR1 基因在胃癌转移中的功能和调控机制,为临床抑制胃癌转移提供新思路。

英文摘要: It is important to clarify the mechanism of gastric cancer metastasis and invasion for clinical therapy and prognosis.It has shown that epigenetic alterations such as the DNA methylation of tumor suppressor gene could lead to gene expression silencing, and then that may enable tumor cells to metastasize or have a selective advantage at the secondary tissue site. We have found that several genes show expression silencing after lymph nodes metastasis by gene chip method.It have been validated that NPR1 in gastric cancer cells shows aberrant DNA methylation in the promoter region by bisulfite sequencing. On the basis of these results, epigenetic mechanism of NPR1 silencing will be analyzed by luciferase assay,demethylation drug treatment and chromatin immunoprecipitation.Simultaneously,NPR1 expression vector is constucted and transfected in gastric cell,then the key role of NPR1 in the metastasis will be determined by clony formation assays,invasion assays and cell migration assays.At last,relation between NPR1 methylation and clinical progression and prognosis will be analyzed.All of these results will clarify NPR1 function and regulative mechanism in the gastric cancer metastasis and provide a new pathway of gastric cancer metastasis inhabition.
