中文摘要: 脑损伤修复是临床亟待解决的重大难题,干细胞对其治疗有极大潜力和重要应用价值。脑损伤后良好的生存环境和诱导条件能最大程度改善干细胞生物学功能,在脑损伤再生修复中发挥关键作用。本课题将利用组织工程方法构建适宜干细胞存活和诱导分化的微环境,探索治疗脑损伤的有效途径。拟将已研制的透明质酸水凝胶基质进一步修饰使之具有保护和诱导神经干细胞的作用,即用具有促进神经干粘附、生长、分化功能的化学功能团和血管内皮生长因子、生长因子、营养因子的活性肽段交联到水凝胶上,建立适于神经干细胞的生存、增殖和分化的局部环境,促进脑损伤修复。在体外细胞水平检测其促进神经干细胞增殖、诱导分化,诱导血管内皮细胞生长的作用;在体内观察对神经干细胞的存活、迁移、分化的作用,评价其对提高脑损伤疗效的程度;探索改善脑损伤疗效的有效途径。

英文摘要: Currently, there are no effective therapies to restore lost brain neurons. Neural stem cells (NSCs) provide promising treatment. A good environment for NSCs survival and differentiation will play key roles for regeneration of central nervous system.Here we design neurogenic properties of bioactive scaffolds to promote survival, proliferation and differentiation of NSCs and repair of brain injury. The scaffolds are generated by amino groups, peptides of VEGF,BDNF,EGF,bFGF and laminin linked to the hyaluronan hydrogel - - the main component of brain extracellular matrix. Neural stem cells are seeded in the scaffolds in vitro to test the effect of the scaffold for adhesion, proliferation and differentiation. Then the scaffolds are transplanted into the rat injuried brain tissue with or without NSCs for repair. We hope that such differentiation resulted in generation of mature neurons and glia cells to replace the dead cells and improve the function. This developed biomaterials may provide new tools for regeneration of central nervous system.
