中文摘要: 磷是动物体内具有重要功能的矿物元素,但由于对磷的吸收和调控机理未完全明确,导致不能准确评定磷的营养需要量、磷的利用率低等问题,并由此引起磷资源的浪费和严重的环境污染。FGF23是最新发现的调节肾脏中磷吸收(Na/Pi-IIa型)的重要因子,但对小肠中磷吸收(Na/Pi-IIb型)的影响未有研究。猪是磷污染排放的主要动物,磷吸收以小肠Na/Pi-IIb为主要载体蛋白,FGF23对其表达和磷吸收是否有影响,其机理如何,未见报道。因此,本项目应用RNA干忧技术、结合动物试验,探明FGF23对猪小肠磷吸收的调控作用,并从三方面探讨其调控机理:①FGF23对维生D转化活性形式过程中相关酶和受体基因表达的影响及其与磷吸收的关系;②对小肠刷状缘囊膜转运磷动力学参数的影响;③日粮中不同磷水平与FGF23、Na/Pi-IIb表达和磷吸收的关系。 项目的完成将为磷的吸收与调控机制奠定新的、积极的理论基础。

英文摘要: Phosphorus is a mineral element which plays an important role in animal metablisms.But because the absorption and regulation mechanism is not entirely clear, the nutritional requirements can't be accurately assessed and lower phosphorus availability.Thereby,it causes serious environmental pollution as well as the waste of resources.FGF23 is a newly discovered hormone protein which regulates phosphorus absorption of renal Na/Pi-IIa, but small intestinal Na/Pi-IIb has not been studied yet. Pigs are the main animals that produce the phosphorus emissions while intestinal Na/Pi-IIb is the most important cotransporter protein for phosphorus absorption,but the effects of FGF23 on it have not been reported. Therefore, this project intends to prove the effects of FGF23 regulation of intestinal Na/Pi-IIb phosphorus absorption by RNA interference(RNAi) combined with the animal experiments,and study the mechanism in the following respects: First, the pathway of FGF23 affect the expression of receptors and enzymes of vitamin D transfering to the active one, and also infer the relationship between that process and phosphorus absorption.
