中文摘要: 蛋白质的N-磷酸化修饰在核小体组装、细胞转录、信号转导等生命过程中发挥了举足轻重的作用。然而由于N-磷酸化肽段对酸敏感,因此采用传统方法难以富集,进而导致了目前国际上对N-磷酸化蛋白组学的研究尚处于起步阶段。为深入了解N-磷酸化蛋白质在生命过程中发挥的作用,亟需发展N-磷酸化蛋白质组样品预处理新方法。本项目拟发展O-磷酸化肽去磷酸化、阴离子交换色谱分级和功能化氧化石墨烯材料富集等新技术,并通过上述技术的集成,建立具有普适性的、高选择性和高效的N-磷酸化蛋白质组样品预处理新方法,为推动N-磷酸化蛋白组的研究进程提供重要的技术支撑。

英文摘要: Protein N-phosphorylation regulates a series of essential functions in life processes, such as nucleosome assembly, cell transcription, signal transduction and so forth. However, traditional methods are difficult to enrich N-phosphopeptides because of its acid-lability, which leads to current international research on N-phosphoproteomics in its infancy. To better understand the roles of N-phosphorylated proteins in life processes, it is urgent to develop new methods for N-phosphoproteome sample pretreatment. The project intends to develop new technologies including O-phosphopeptide dephosphorylation, anion exchange chromatography fractionation and functionalized graphene oxide enrichment, and establish a universal, high selectivity and high efficiency approach for N-phosphoproteome sample pretreatment by integrating the above technologies, which will provide important technical support for promoting the process of N-phosphoproteome research.
