中文摘要: 脂蛋白相关磷脂酶A2(Lp-PLA2)是预测心血管疾病发生和粥样斑块不稳定性的炎症相关因子。在人体循环中主要与LDL结合,促动脉粥样硬化发生;而在小鼠中,却主要与HDL结合,对心血管起到保护作用。与小鼠相比,猪的脂蛋白谱与人类相似,是研究Lp-PLA2相关机制的最适模型。在本实验室前期制作的人载脂蛋白C3(apoCIII)转基因猪血液中,Lp-PLA2的活性是对照组的3倍,预实验结果显示,apoCIII在平滑肌细胞中显著上调Lp-PLA2的表达。因此,本研究拟以apoCIII转基因猪为基础,以Lp-PLA2为研究对象,在猪胸主动脉和平滑肌细胞中,用分子和细胞生物学方法检测apoCIII对Lp-PLA2的调节和对其介导平滑肌细胞生物学行为的影响;并采用启动子和信号通路分析等方法深入阐明apoCIII调节 Lp-PLA2的分子机制,为研究Lp-PLA2在动脉硬化发生发展中受调节的机制奠定基础。

英文摘要: Lipoprotein-associated phospholipase A2 has been considered as an independent inflammatory biomarker of both coronary heart disease and unstable plaque which can predict and regulate the development of atherogenesis.Lp-PLA2 is associated primarily with LDL in human and promote atherosclerosis; whereas,in mice, Lp-PLA2 is predominantly associated with HDL and exhibit antiatherosclerotic effect. But porcine has lipoprotein profile similar with human and is considered the suitable model to Lp-PLA2 study. In the circulation of apoCIII transgenic pig which our lab did previously, Lp-PLA2 activity is three fold compare with the nontransgenics';In addition,apoCIII can upregulate Lp-PLA2 in porcine smooth muscle cells. Therefore, based on the apoCIII transgenic pig, we examine the regulation of Lp-PLA2 levels with apoCIII and Lp-PLA2 mediated biological changes of vascular smooth muscle cells with molecular and cellular techniques; promoter and signalling pathway analysis will use to confirm the molecular mechanisms of the regulation of Lp-PLA2 with apoCIII deeply. The results may contribute to the regulated mechanisms of Lp-PLA2 in atherogenesis.
