中文摘要: 研究开发和利用能源植物是国家和云南省产业发展战略的需求,小桐子(Jatropha curcas L.)是具有巨大开发潜力的能源植物树种。探索植物油脂的生物合成与调控,以提高种子含油量一直是小桐子育种的重要任务之一。在项目组前期分离了小桐子FAD, ACP等个7结构基因的基础上,本项目拟以两组(各3个)含油量不同的小桐子种群为材料, 开展下列研究:①两组含油量不同的小桐子种群种子发育过程中转录组的变化与比较分析;②与小桐子油脂生物合成调控相关关键转录因子的时空表达分析及完整CDS的克隆;③种子特异表达启动子驱动的过表达载体的构建,转化烟草和小桐子,进行基因功能解析和获得转基因小桐子。本研究成果将在一定程度上揭示小桐子油脂生物合成调控的分子机制,克隆出与油脂生物合成调控相关关键基因,并进行功能解析。研究成果具有重要的科学意义和潜力巨大的应用前景。

英文摘要: Research and development of energy plants is a strategical demand for Yunnan province and our country. Jatropha curcas L. is a sustainable energy plant with great potential for biodiesel production. In order to improve the seed's lipid content, one of the important tasks of Jatropha curcas breeding is to research the lipid biosynthesis and regulation. Based on cloned 7 structural genes such as FAD, ACP, using two populations of J. curcas with different lipid content as experimental materials, the following items are to be studied in this proposal: ① changes of transcriptomes and their analysis and comparison in J. curcas seed during seed development. ② analysis of saptio-temporal expression pattern of those key ESTs correlated with lipid biosynthesis regulation and cloning those key genes correlated with lipid biosynthesis regulation. ③Construction of seed specific promoter expression vector, and transformation into Nicotiana tabacum and J. curcas, functional characterization of those key genes correlated with lopid biosynthesis regulation, genetic transformation of J. curcas using the above-mentioned key genes correlated with lipid biosynthesis regulation to enhance their lipid biosynthesis. The research will elucidate molecular mechanism of lipid biosynthesis regulation, clone some key genes correlated with lipid biosynthesis regulation, and analyze the functional characterization of those genes. These results will be of scientific importance and great potential for application.
