中文摘要: 接种根际微生物是提高超积累植物对土壤中重金属修复效果的重要途径,但目前对根际微生物的耐受与修复机理还不完全清楚。本研究拟从重金属耐受性和溶解性两方面进行筛选,在Cu、Zn污染土壤中筛选并鉴定出具有高Cu、Zn耐受性和溶解性的微生物菌株,研究Cu、Zn在微生物亚细胞的分布及化学形态,分析Cu、Zn在微生物细胞中的累积特点和结合形式,通过微流控单细胞分析、抗氧化系统毒性评价与促植物生长指标研究,从细胞水平全面阐明微生物对Cu、Zn耐受性与促植物修复的发生机制,最后,对Cu、Zn富集植物酸模进行微生物接种,分别通过温室盆栽和试验田测试,分析微生物菌株对酸模修复土壤中Cu、Zn的促进效果,评价微生物菌株用于实际土壤Cu、Zn修复的应用潜力。本课题有助于全面认识微生物菌株对土壤中Cu、Zn的耐受与修复机理,酸模结合筛选的菌株对土壤Cu、Zn的修复具有潜在的应用价值。

英文摘要: Inoculation with rhizosphere microbes is an important way to improve restoration effects of hyperaccumulators on heavy metals in soil. However, at the present time, the tolerance and restore mechanism of rhizosphere microbes has not been fully understood. This research will screen microbes from heavy metal tolerance and solubility. Microbial strains with high Cu, Zn tolerance and solubility will be screened and identified in Cu, Zn contaminated soil. Microbial subcellular distribution and chemical forms of Cu, Zn will be studied. Cu, Zn accumulation features and combined forms in microbial cells will be analyzed. Through microfluidic single-cell analysis, toxicity evaluation of the antioxidant system and bio-stimulant indicators, the mechanism of microbial Cu, Zn tolerance and promoting phytoremediation will be fully clarified from cellular level. Finally, the Cu, Zn hyperaccumulator Rumex Acetosa will be inoculated with microbes. Through the greenhouse and experimental field test, the promoting effects of microbial strains on Cu, Zn restoration by Rumex Acetosa will be analyzed. Further, the application potential of microbial strains used for Cu, Zn restoration in actual soil will be evaluated.
