中文摘要: 采用不同培养基质的选择性分离培养方法,对河西走廊地带原发盐碱地、次生盐碱土壤环境中的可培养放线菌进行分离,通过形态特征、耐盐碱性实验、抑菌实验等研究探讨这一特殊生境中可培养放线菌的主要类群和分布特征,建立河西走廊放线菌种质资源库;采用非培养的基于16S rRNA基因序列系统发育分析方法进行比较研究,结合传统培养技术探讨河西走廊盐碱环境中放线菌分布规律和物种多样性,阐明该地区放线菌与土壤理化因子的关系。本研究及其结果对甘肃河西走廊盐碱环境下放线菌物种多样性和放线菌生态研究起到填补和推动作用,促进对该地区放线菌种质资源的保护,为盐碱环境放线菌在筛选新抗生素及工业微生物酶制剂等领域的开发研究及河西走廊盐碱土壤的综合改良提供基础资料。

英文摘要: Using selective screening method with different selective medium, to isolate actinomyces in the primary saline-alkali soil and the secondary saline-alkali soil of Hexi Corridor in Gansu Province. By the morphological characteristics, alkali experiment and antibacterial experiment, the cultivable class group and distribution characteristics of actinomyces in this special habitat are investigated, and the actinomycetes germplasm bank is set up. Based on 16 S rRNA gene sequence analysis method of phylogenesis and traditional culture method, the regularity of distribution and diversity of actinomyces in saline-alkali environment of Hexi Corridor are discussed, the ecology characters of actinomyces in this region is clarified. The results of this research play an important role to the study of actinomycetes diversity and actinomycetes ecology of the saline-alkali environment in the Hexi Corridor, especially provide a reliable basis data for actinomyces resource protection in this region, and provide the basic data in selecting new antibiotics, industrial microbial enzymes and other areas of saline-alkali environment, and provide the basic data to multiple improve the saline-alkali soil of Hexi Corridor.
