中文摘要: 从福建红树林筛选,并通过选育得到一株齿毛菌(Cerrena sp.)HYB07菌株,该菌株所产碱性耐盐漆酶,性质稳定独特,在海洋污染物降解方面有广阔的应用前景。本课题为应用基础研究,拟就以下几方面开展研究:1)系统研究HYB07菌株中漆酶家族9个成员的基因序列与蛋白、酶学性质,以便选择性质优良的漆酶进行高效表达和应用研究;2)结合漆酶基因启动子的生物信息学分析和基因转录谱,考察漆酶基因的转录调控机制,服务于后续发酵产酶及应用条件优化;3)研究漆酶同工酶在降解偶氮、蒽醌、靛类、三苯甲烷、多环芳烃等化合物方面的效果、产物和机制,建立其在海洋污染防治方面的核心技术体系;4)构建漆酶的高效表达体系,为其下一步的研究、生产和应用奠定基础。

英文摘要: In search of new laccases with desired properties, a novel laccase-producing fungal strain, Cerrena sp. HYB07, was isolated from a mangrove swamp in Fujian Province and bred selectively. Laccases from HYB07 are alkaline, halotolerant and stable over a wide range of pHs and temperatures and therefore may have broad applications in treatment of marine pollutants. The proposed project aims to study the HYB07 laccase family in the following aspects: 1) cloning of the nine laccase genes, their heterologous expression in yeast Pichia pastoris and analysis of the enzymatic properties of the recombinant proteins; 2) with detailed promoter analysis and transcription and enzyme profiling, transcription regulation of the laccase genes will be elucidated for subsequent optimization of fermentation conditions and their applications; 3) studying the degradation and detoxification effects of the laccases on azo, anthraquinone, indigo, triphenylmethane and polycyclic aromatic hydrocarbon compounds, identifying the products and the underlying mechanisms of action as well as establishing core technical parameters for laccase-mediated marine pollution control; 4) realizing high-level expression of the laccase(s) with desired enzymatic properties and application potential.
