中文摘要: 运动性低血睾酮是运动员体能低下的重要因素,成为制约运动员运动能力正常发挥的关键问题。睾酮只有与其受体-雄激素受体(androgen receptor, AR)结合后才能发挥生理功能。本课题一方面研究不同强度的耐力运动、牵拉刺激分别对大鼠腓肠肌和比目鱼肌、小鼠骨骼肌细胞C2C12中AR表达和活性的影响及其机制-是否与雄激素、MAPK通路(p38、ERK、JNK)、Akt通路(PI3K、Akt)和胰岛素样生长因子1(IGF-1)/胰岛素样生长因子1受体(IGF-1R)有关;另一方面研究骨骼肌AR的蛋白水平和活性对大鼠运动能力的影响。在此基础上,研究蒺藜提取物提高过度疲劳运动大鼠运动能力的机制,即是否是通过MAPK、Akt和IGF-1/IGF-1R提高骨骼肌中AR的表达和活性。这些研究结果不仅加深了AR与运动关系的理论认识,还为开发蒺藜提取物为提高过度疲劳运动后运动能力的运动饮品提供理论依据。

英文摘要: High level of serum testosterone is beneficial to increase and keep exercise ability of athletes. Testosterone plays its biological effects through the mediation of androgen receptor (AR). In this paper we will study the following aspect. First, determine the protein level of AR and its activity of activated transcription in soleus and gastrocnemius of rats after different exercise intensity and in C2C12 cells after different intensity stretch to explore the effect of exercise on AR expression and activity, then clarify if the effects of exercise on AR are associated with androgen level, MAPK pathway (p38、ERK、JNK), Akt pathway (PI3K、Akt) and insulin-like growth factor 1 (IGF-1)/ insulin-like growth factor 1 receptor (IGF-1R). Second, investigate the influence of the protein level of AR and its activity in soleus and gastrocnemius of rats on the activity performance. Last, explore the improved role of Tribulus terrestris on activity performance of over-trained rats, and detect if the mechanisms are related to MAPK pathway, Akt pathway and IGF-1/IGF-1R.
