中文摘要: 阿尔茨海默病(AD)是老年人群健康的主要威胁之一。本课题前期临床研究发现,轻中度AD患者以心气虚证为主,且调心方能够明显提高患者的认知功能,但机制尚未阐明。越来越多的证据表明, 物质能量代谢紊乱促进AD的发生发展,NAD+-SIRT1能量通路在AD病理损伤的早期阶段发挥着重要作用。本项目将探讨NAMPT/NAD+/SIRT1/PGC-1α信号通路以及miR-34a在早期AD发病进程中的参与机制,探讨调心方在其中作用的靶点。论证调心方可否通过直接或通过miR-34a上调NAMPT的表达来改善能量代谢紊乱,以期该研究能解析调心方干预早期AD病变的效应靶点和分子机制,为中医药防治AD提供新的实验室依据。

英文摘要: Alzheimer's disease (AD) is one of the major threats of aging population. The preliminary clinical studies have found that: AD patients with mild to moderate disease usually suffer Heart-qi deficiency; Tiaoxin Recipe can significantly improve the cognitive function. But the mechanisms underlying the role of Tiaoxin Recipe in intervening AD remain largely unknown. M accumulating evidence show that energy metabolism disorder promotes the development of AD, energy pathway of NAD+-SIRT1 play an important role in the early stages of AD. This project will set out to investigate the mechanism underlying the expression of NAMPT/NAD+/SIRT1/PGC-1α signaling protein and miR-34a in the early stage of AD, and to explore the mechanisms of Tiaoxin recipe, and to eventually answer the question whether Tiaoxin Recipe could improve metabolic disorders via increasing the expression of NAMPT or inhibiting the expression of miR-34a. Our study will identify the effective target of Tiaoxin Recipe and elucidate the molecular mechanism by which Tiaoxin Recipe intervening mild to moderate AD, and to provide experimental evidence for the treatment and prevention of AD by traditional Chinese medicine.
