中文摘要: 染色质组装因子CAF-1是一种真核生物中高度保守的组蛋白分子伴侣,含有三个亚基。它的一个主要作用是在DNA复制过程中将新合成的H3.1-H4异二聚体组装到正在复制的DNA上。除此之外,CAF-1还参与了细胞的DNA损伤修复和基因沉默等过程。因此,CAF-1是基本生命过程中非常重要的蛋白复合体。自发现二十年来,关于CAF-1的功能研究有很多,但关于CAF-1的作用机制仍所知甚少。我们希望通过研究CAF-1各个亚基的结构与功能,特别是大亚基的功能,以及各个亚基之间的相互作用关系来研究CAF-1的组装及分子机制。我们初步的实验结果研究了各亚基的精细相互作用关系,发现了酵母中CAF-1具有直接结合DNA的能力,而且在CAF-1大亚基Cac1中发现一个能够结合DNA的结构域。我们希望通过生化、细胞生物学及结构生物学等手段来进一步揭示各亚基的结构﹑功能以及分子机制。

英文摘要: CAF-1(chromatin assembly factor 1), consisting of three subunits, is a highly conserved histone chaperone in eukaryotic cells,which assembles newly synthetic histone H3.1-H4 heterodimer to nascent DNA during DNA replication. CAF-1 also functions in DNA repair and heterochromatin silencing. In the past twenty years, many studies about CAF-1 have been reported, however the functional mechanism of CAF-1 is still unclear. We plan to study CAF-1 by studying the structures and the function of its three subunits and the interactions among them. Our recent preliminary results revealed the fine interaction of the subunits, and found that yeast CAF-1 can bind DNA, which is similar to another histone chaperone Rtt106. A DNA binding region was further identified within the large subunit Cac1. We will further the study the interactions and the functions of the three subunits by structural and cellular approaches, in order to reveal the mechanisms underlying this important protein complex.
