中文摘要: 以解决生物医学研究中纳米颗粒与细胞相互作用的问题为目标,课题创新性地提出利用光学标测技术在细胞层级实现纳米颗粒与细胞膜相互作用的光学表征方法。依据细胞膜性质和结构的变化会引起细胞膜电位的变化,结合细胞电生理学和传质学理论,建立细胞膜电位变化和纳米颗粒与细胞膜作用关系的数学模型,表征其与膜上电位变化的关系,为纳米材料在生物医学领域的应用研究提供评价标准。 .同时利用该方法,构建适合细胞尺度研究的光学标测系统,采用新技术新手段探讨不同性质的纳米颗粒与细胞膜的相互作用机制。从不同粒径、不同表面带电性以及不同表面修饰环境的金纳米颗粒中寻找跨膜性强同时对细胞膜损伤低的纳米颗粒表面修饰方式,研究和表征不同性质金纳米颗粒与细胞膜作用机制。推动纳米颗粒在生物医学领域的研究进展,为研究纳米颗粒与细胞相互作用提供一种崭新的技术手段。

英文摘要: To solve the current problems about nanoparticle-cell interaction in biomedicine, we plan to use new technologies and new methods to explore the mechanism of anoparticle–cell membrane interaction. Because membrane properties and structural changes will cause changes in membrane potential, we can build up a mathematical model to describe and characterize the relationship between changes in membrane potential and the nanoparticle–cell membrane interaction based on the theory of cell electrophysiology and mass transfer study, and promote a new evaluation criterion for nanomaterials application research in biomedicine..Based on this method, we will build cell-level optical mapping system and discover the mechanism of .the interaction of gold nanoparticles with different properties with cell membrane. Looking for the optimized surface modified method of nanoparticles by considering the effect of gold nanoparticles with different size, different surface charging and different surface modified environment to promote nanoparticle drug applied research progress. And also provide a novel technical method for the study of nanoparticle–cell interaction.
