中文摘要: 近年来,Q烟粉虱及其传播的番茄黄化曲叶病毒(TYLCV)病在我国持续暴发危害,申请者前期研究发现,番茄感染TYLCV后,其挥发物质有很大改变,导致Q烟粉虱喜欢选择感染TYLCV的植株取食。因此,我们推测番茄挥发物是促进Q烟粉虱及其传播的番茄黄化曲叶病毒病大暴发的重要原因。昆虫气味结合蛋白(OBP)可以和脂溶性的寄主植物挥发物结合,在昆虫与寄主植物互作中起着重要作用。为明确Q烟粉虱对番茄植株挥发物的识别及感受机制,本研究将测定Q烟粉虱对番茄挥发物标样的嗅觉反应,并克隆鉴定三个OBP,制备目的基因的dsRNA,利用RNAi技术,测定目的基因沉默后,Q烟粉虱对番茄挥发物的反应特性。揭示Q烟粉虱OBP在TYLCV传播中的功能机制。研究结果对于明确Q烟粉虱嗅觉感受机制、丰富烟粉虱-寄主植物-双生病毒间的互作理论具有重要意义,并为开发Q烟粉虱趋避剂、促进Q烟粉虱及其传播的病毒病可持续防控提供科学依据。

英文摘要: In recent years, the spread of Bemisia tabaci Q and tomato yellow leaf curl virus (TYLCV) disease outbreak continued in the country, applicants preliminary found that after infected TYLCV, the volatile substances of tomato have changed dramatically, and it is resulted in Bemisia tabaci Q preferring to feed on TYLCV infected tomato plants. Therefore, we hypothesized that the tomato plant volatiles is an important reason to promote the spread of Bemisia tabaci Q and TYLCV disease outbreak. Insect odorant binding protein can combine with fat-soluble of Host plant volatiles, which plays an important role in the interaction between insects and host plant. In order to clarify the mechanism of Bemisia tabaci Q identification and feel to tomato plant volatiles, this study will measure the olfactory response of Bemisia tabaci Q to tomato volatiles standards, clone and identify the three odorant binding proteins, preparation dsRNA of receptor gene, and use RNAi technology, determinate the reaction characteristics of Bemisia tabaci Q to tomato plant volatiles after the receptor gene silenced. Reveal the function mechanisms of odorant binding proteins of Bemisia tabaci Q on the transmission of TYLCV. This findings will have important significance for clearing the mechanism of Bemisia tabaci Q olfactory receptor, enriching the interaction theory between whitefly-host plant-geminiviruses, it will also provide a scientific basis for the development of Bemisia tabaci Q refused, promote the sustainable prevention and control of Bemisia tabaci Q and the disease transmitted by it.
