中文摘要: 慢性炎症与食管鳞癌发生密切相关,炎性相关基因表达失调是其中关键的生物学事件。然而,食管鳞癌中炎性相关基因表达失调与个体表达差异的分子机制仍未阐明。以往的工作要么针对单个基因,要么针对遗传或表遗传单一机制研究,未能全面解释其真正机制。本项目运用高密度芯片与新一代测序等技术平台,系统研究食管癌组织中表达失调的炎性相关基因的遗传变异、拷贝数变化、DNA甲基化及miRNA变化与基因表达的关系,构建炎性基因表达失调的分子调控网络,从遗传学和表遗传学整体上阐明食管鳞癌发生发展中炎性相关基因表达异常的机制,为食管鳞癌的预防、个体化治疗和发现药物和治疗靶点提供新思路。

英文摘要: Chronic inflammation is implicated in the pathogenesis of esophageal squamous-cell carcinoma (ESCC), and the aberrant expressions of inflammatory genes are critical events in the development of ESCC. However, the underlying molecular mechanisms for the deregulation and individual variation of inflammatory gene expression in ESCC are largely unknown. In this study, we will systematically examine the effects of genetic variations, copy number alteration, DNA methylation and/or miRNA alteration on the expression of inflammatory genes in ESCC tissues, and then integrate the genome-wide data from different technologies into the molecular regulatory network of inflammatory gene expression in ESCC. The results of this study will provide new insights into the mechanisms of deregulation of inflammatory genes in ESCC, which has profound significance for the prevention and personalized therapy of the cancer.
