中文摘要: 本项目旨在利用聚合物蛋白质耦合体为构筑基元探索构建多室蛋白质囊泡的形成机制及合成方法并实现对内室数目和尺寸的调控。进而在此基础上通过靶向引入多酶串联体系到该多室蛋白质囊泡的指定区域,实现多室多酶蛋白质囊泡体系的构建并研究其在细胞自维持等性质的人工模拟等方面应用及酶空间位置的调控对该体系协同催化效率的影响。近年来,人工细胞模型的构建引起了来自不同领域科学家们的广泛关注,该领域的发展一方面可以加强我们对细胞的进一步认识,探索其进化规律为可能的生命起源提供新的视角;另一方面构建的模型也展现出了在蛋白质、RNA 及药物分子的体内输送或在生物反应器等方面应用的巨大潜能。其中,最近由申请人首次报道的单室蛋白质囊泡作为新型人工细胞模型被同行高度评价。在此基础上,本项目拟建立的多室多酶蛋白质囊泡体系模拟一些更复杂的细胞行为,将被视为下一代人工细胞模型,为该领域的发展贡献新的力量。

英文摘要: In this project, we aim to establish a new method to construct multi-compartment proteinosomes with controlled number of inner compartment and size. Then, by targetingly incorporating three cascaded enzymes into different domains of the multi-compartment proteinosome, the constructed multi-compartment/enzyme proteinosome, as a novel protocell model, will demonstrate a potential application on mimicking parts of cell properties such as self-maintenance behavior. Recently, more and more researchers from different communities paid great attention on the field of artificial cell, given that on one hand it will help us to understand the evolution of cell and bring another insight to the origin of life, on the other hand the models show promising application in the fields including protein/RNA/drug delivery or artificial bioreactor. Significantly, among these, the proteinosome reported by the applicant as a novel protocell model was highly recognized very recently. Herein, continuing this work, this project is aiming to further construct multi-compartment proteinosome to mimic some advanced cell properties. Undoubtedly, such multi-compartment proteinosome to be the next generation model, will contribute the development of this field greatly.
