中文摘要: 宫颈癌放疗中的辐射抵抗是影响临床放疗效果的重要因素,而发生辐射抵抗的机制尚不明确。因此,研究影响宫颈癌辐射敏感性的因素,探索其机制,具有重要的临床意义。肿瘤血管生成是影响辐射敏感性的重要因素之一,HIF-1α是公认的影响肿瘤辐射敏感性的重要基因。既往研究发现,肝素酶1既能促进肿瘤血管生成,又能上调HIF-1α的表达。我们的前期研究表明,在宫颈癌HeLa细胞中,上调HPSE1可诱导辐射抵抗,且宫颈癌放疗后未控或复发患者的肿瘤组织中HPSE1高表达。我们推测肝素酶1通过促进肿瘤血管生成、上调HIF-1α的表达介导宫颈癌辐射抵抗。本项目拟通过临床宫颈癌标本、体外实验和荷瘤鼠体内实验,在不同的肝素酶1表达水平下,研究肿瘤血管生成情况及相关分子表达,HIF-1α的表达以及辐射敏感性,并通过干预相关信号通路,明确肝素酶1在宫颈癌辐射抵抗中作用,并研究其分子的机制,为临床宫颈癌放疗提供新的增敏策略。

英文摘要: Radioresistance in the process of treatment of cervical carcinoma is principal element which was impact on the therapeutic benefits of radiotherapy. The mechanism of radioresistance is still unknown. Therefore, it is very important to find the influencing factor and mechanism of radiosensitivity of cervical carcinoma. Tumor angiogenesis is important element of radiosensitivity. Angiogenesis factor as HIF-1α, VEGF is also important to radiosensitivity. Heparanase 1 (HPSE1) has impact on tumor angiogenesis, invasion, and metastasis. Recent studies show that HPSE1 influence the expression of HIF-1α and VEGF. The result of our preliminary studies shows that the radiosensitivity decreased in HPSE1 highly expressed HeLa celllines, and that the tumor tissue of cervical carcinoma which was radiioresistance expressed high level of HPSE1. We infer that HPSE1 mediate radioresistance through tumor angiogenesis. This project will study the relation of HPSE1 and radiosensitivity with clinical tissue of cervical carcinoma, in vitro and in vivo studies. We make further study of interact factors of HPSE1 with coimmunoprecipitation and mass spectrometry. The functional test will be made to explore the mechanism of HPSE1 mediate radioresistance and to find a novel strategy of hypersensitivity in radiotherapy of cervical carcinoma.
