中文摘要: 本项目研究了多频超声波处理促进蛋白质酶解反应的机理及控制机制。与单频超声处理、未经超声处理比较,本研究重点评价多频超声处理的酶解效果,优化多频超声处理的工作模式和主要参数;建立基于多频超声波处理的酶解反应动力学和热力学模型;探讨蛋白质和蛋白酶波谱特性、结构特征改变与多频超声波处理促进酶解反应之间的内在关联;从化学反应动态学和分子水平两个层面阐明多频超声波处理促进蛋白质酶解反应的机理,建立多频超声促进蛋白质酶解反应的控制机制。该项目不仅可以突破传统蛋白质酶解制备功能性多肽技术的发展瓶颈,而且可以推进超声波新技术从常规工作模式到高级工作模式的发展,也将进一步推动农产品与食品精深加工领域及健康产业的发展。

英文摘要: The protein enzymolysis reaction promoted by multi-frequency ultrasound and its mechanism were studied in this project. Compared with the mono-frequency ultrasound processing and the processing without ultrasound, the enzymolysis performance under the multi-frequency ultrasound processing was systematically evaluated. The work mode and main parameters of multi-frequency ultrasound processing were further optimized. Moreover, the reaction kinetic and thermodynamic models of enzymolysis under the multi-frequency ultrasound processing were established. The internal relations between the spectral characteristics and architectural features of protein and protease and the enzymolysis reaction promoted by multi-frequency ultrasound were discussed. From the chemical reaction dynamics and molecular level, this research illuminated the protein enzymolysis reaction mechanism promoted by multi-frequency ultrasound and developed corresponding control mechanism. The proposals of this project were to resolve the development bottlenecks of traditional enzymolysis technology for functional polypeptides preparation from protein, promote the development of new ultrasonic technology from conventional work mode to advanced one, and push the deep-processing field in agricultural products and food and the health industry forward.
