中文摘要: 本课题在已获得猪杂交亲代与子代和中外猪种肌肉组织差异表达基因的基础上,通过对差异表达基因CKM(骨骼肌能量代谢关键酶)、MLC2(在肌肉发生过程中起着调控重链表达的重要作用)和ITGB1BP2(在肌细胞成熟和组织形成过程中发挥重要作用)启动子的克隆、鉴定、甲基化分析和功能性多态位点筛查,鉴定了CKM、MLC2和ITGB1BP2的核心启动子区域和转录起始位点以及启动子区不同缺失片段的活性和与启动子相结合的转录因子MEF-2、MyoD的调控作用;研究了不同品种、不同阶段及不同组织启动子区CpG岛甲基化状态以及与基因表达的关系,表明基因表达与CpG甲基化程度呈负相关;分析了启动子区多态位点与猪经济性状的关系以及对基因表达的影响,获得有效分子标记4个。除此之外,还研究了猪肌肉基因表达分析的内参基因以及猪Six1和Pitx2c 基因。到目前为止,申请专利6项,发表SCI收录论文3篇,会议论文2篇,在线发表论文2篇,取得了阶段性的研究成果,达到了预期目标。

英文摘要: Based on the isolation of differentially expressed genes between the muscle tissues of pig hybrid and their parents or foreign breed and local breed, we cloned the promoters of differentially expressed genes CKM (a crucial enzyme in energy metabolism of skeletal muscle), MLC2 (which play an important modulatory role in myosin heavy chain expression during muscle development), ITGB1BP2 (which play a crucial role during maturation and/or organization of muscle cells), identified their core promoter, transcription start site, activity of promoter fragment and regulation of transcription factors MEF-2, MyoD that bind to the promoters, analyzed the relationship between methylation level and gene expression, investigated the effect of functional polymorphism loci in promoter on pig economic traits and gene expression. In addition, we also identified reference genes for gene expression studies in porcine skeletal muscle, and studied porcine Six1 and Pitx2 gene. Up to now, 6 patents were registered, 3 original articles and 2 conference papers were published, and 2 papers were published online. Some ideal results have been obtained. The results have an important academic value and extensive application perspective.
