中文摘要: 染色质的长距离相互作用与基因转录调控有着密切的关系,然而在细胞分化过程中大规模染色质相互作用的变化规律及其分子机制仍有待研究。在初始T细胞分化成Th2细胞过程中,许多基因位点上的表观遗传修饰以及基因转录水平都发生了明显变化,为系统性研究染色质长距离相互作用提供了良好的研究体系。本项目拟采用最新的全基因组规模的染色质相互作用研究技术- - ChIA-PET,与计算生物学分析相结合,系统性鉴定Th2细胞中基因启动子与增强子间长距离的相互作用;并将其与分化前初始T细胞相比较,找出分化过程中染色质高级结构的变化规律和相应基因转录变化的相关性。同时,本项目将进一步研究Th2细胞分化过程中GATA3等重要因子对于染色质相互作用的调控功能与分子机制。本项目不仅有助于进一步阐释染色质高级结构对于基因转录的调控功能,而且对于深入理解免疫细胞分化的调控途径和分子机制也会提供重要帮助。

英文摘要: Long-range chromatin interactions are involved in gene transcriptional regulation. However, the detailed mechanisms of how chromatin interaction patterns change during cellular differentiation process are largely unknown. Th2 cell differentiation is a dynamic process with significant changes of epigenetic modifications and gene transcription levels on many loci, thus providing an ideal system to study long-range chromatin interactions. This project is aiming to systematically study the long-range chromatin interactions between gene promoters and enhancers in Th2 cells with the state-of-art technology, ChIA-PET. In particular, this project will identify the changing patterns of chromatin interactions during Th2 cell differentiation and establish the correlations between chromatin interactions and gene transcription. Further, this project will investigate the molecular mechanisms of the regulatory roles of some important factors such as GATA3 in mediating long-range chromatin interactions in Th2 cells. Our project will not only help us to better understand the mechanisms of how higher-order chromatin structure regulates gene transcription, but also provide great insight into regulatory pathways in the differentiation process of immune system.
