中文摘要: 丹参酮类有效成分是中药丹参中一类脂溶性二萜醌类化合物,目前对其代谢途径的关键酶研究颇多,但对其途径的调控基因研究较少,这严重制约了对丹参酮类化合物形成及其调控的全面解析。长链非编码RNA是一类转录本长度超过200 nt的RNA分子,它们并不编码蛋白,而是以RNA形式在表观遗传、转录以及转录后调控等层面上调控基因的表达。为获得丹参长链非编码RNA以及探讨该类基因是如何参与调控丹参酮类化合物的形成,本项目拟通过构建丹参转录组文库,对转录组数据分析获得丹参长链非编码RNA,运用过表达和RNAi技术,构建长链非编码RNA过表达和表达抑制载体,分别转化至丹参毛状根中,结合丹参酮类化合物形成和积累的变化,阐明丹参长链非编码RNA在丹参酮类化合物形成中的作用及其调控机制。本项目的研究对于揭示丹参酮类化合物形成的分子机制具有重要意义,为保护我国药用植物丹参的重要基因资源及其可持续利用研究提供科学依据。

英文摘要: Tanshinones, the major lipophilic bioactive components of Danshen, are abietane-type diterpenoids. Previous attempts mainly focused on the key enzymes of the biosynthesis pathway, while the regulatory elements received little attention, which is the major bottleneck in comprehensive analysis of the formation and regulation of tanshinones. Long non-coding RNA (lncRNA), a class of RNA molecules longer than 200nt, does not encode proteins, but take part in the epigenetic, transcriptional and post-transcriptional regulation of gene expression. In order to obtain and identify the transcriptional regulators involved in the biosynthesis of tanshinones, in this project, we intends to use transcriptome profiling of the hairy root of Salvia for identification of lncRNA regulating tanshinone synthesis in Salvia. By over-expression and siRNA-mediated gene silencing of lncRNA in the hairy root of Salvia, and quantification of tanshinones,with dCHIRP analysis we will clarify the role of lncRNA in the formation of tanshinones and the regulation mechanism in Salvia. As a science projection, its harvests would reveal the molecular mechanism of tanshinone formation and significantly provide a firm foundation for the protection of important genetic resources of medicinal plants Salvia and its sustainable utilization.
