中文摘要: 量子点是具有特殊光电性能的功能型半导体纳米材料,在生物医学方面应用前景广阔,其生物效应及毒理学研究已成为当前亟待解决的前沿课题之一。本项目拟以功能化水溶性CdTe量子点为研究对象,采用微量热、荧光、电化学、显微等方法,研究不同粒径、不同表面修饰和不同壳层结构的量子点与酿酒酵母细胞相互作用及产生生物效应的机制。获取相关作用的热力学和动力学参数,并探讨量子点粒径、表面修饰、结构和电荷等因素对其生物效应热力学和动力学性质的影响、及其规律性。结合电镜、显微、XPS、MTT、ICP-MS等,从微观结构诠释所得规律。从细胞代谢热动力学、形态、呼吸功能、摄取量子点方式及其对细胞器的损伤等方面,探讨量子点生物效应的成因,揭示量子点对生命代谢动态过程中的影响途径。本研究将为量子点的高效制备、应用及生物安全性评估,提供理论基础,并且成为纳米材料生物效应研究的模型。

英文摘要: As a novel nanomaterial, quantum dots (QDs) are currently attracting great interest in almost all fields due to their unique electric and optical properties, especially in the fields such as biomedicine. Its biologic effect and toxicity is solved as one of the topics at the forefront. The purpose of this project is to investigate the biologic effect and its mechanism induced by the multiple functionalized CdTe QDs with different sizes, different coatings and different shells. Thermodynamic and kinetic parameters will be determined, and influences of QDs size, surface structure as well as compositions of the media on the thermodynamic and kinetic properties of biologic effect will be investigated. Some useful rules will be summarized and explained based on electronic microscope observation and analyses of spectra of fluorescence, XPS, MTT、ICP-MS etc. Also, factors of biologic effect caused by QDs or other nano materials are further explored from the perspective of cellular metabolism, morphology, respiration fuction, uptake and localization. It will promote us deeply understand the process of effect of QDs on living dynamic things. The research will provide methodology and theoretical basis for the efficient synthesis, application and biosafey evaluation of quantum dots and the modle of toxicity of nanoparticles research.
