中文摘要: MYB转录因子家族成员参与植物发育和代谢等多种生理过程,在调节次生细胞壁发育中具有重要的作用。本研究在获得了白桦次生细胞壁发育相关MYB转录因子BpMYB2基础上,分别在白桦中过表达和抑制表达BpMYB2,分析转基因和野生型植株之间次生细胞壁形态,纤维素、半纤维素和木质素量等性状的差异,确定BpMYB2对白桦次生细胞壁形成的影响;通过转录组学分别比较转基因和野生型植株间的基因表达差异,筛选BpMYB2所调控的下游靶基因,在白桦基因组信息的基础上克隆重要下游靶基因启动子,利用染色质免疫共沉淀(ChIP)技术进一步验证。利用MEME软件分析BpMYB2所调控的下游靶基因启动子的保守序列,进而筛选出BpMYB2识别的顺式元件,并利用EMSA等技术进一步验证。通过对BpMYB2所调控的下游靶基因及识别元件分析阐明其调控次生细胞壁发育的分子途径。为利用分子生物学手段培育优良白桦材新品种奠定基础。

英文摘要: MYB transcription factors are involved in development and metabolism processes of plants,and play important roles in secondary cell wall synthesis of palnts. Based on the fundation of cloning a BpMYB2 gene that is found to related to secondary cell wall development from Betula Platyphylla. The transgenic plants overexpressing or inhibit expressing BpMYB2 gene will be generated in this project, and the differences of cell wall shapes and chemical contents between the transgenic plants and control plants are analyzed.The transcriptoms of the transgenic plants and control plants are bulit using the high throughput sequencing method. Then the down-stream target genes of BpMYB2 will be determined by comparinsing the differentially expressed genes between overexpression and inhibit expression transgenic palnts. The promoters of the target genes are cloned base on the genomic information of B.platyphylla available now. Moreover, some target genes will be selected for confirmation by ChIP (chromatin immunoprecipitation assay) method. The motifs of the target gene promoters are searched using MEME sofeware, and the conserved seuquences among these promoter will be identified. The conserved seuquences identified by MEME may bind to the MYB, and the specific binding between BpMYB2 and the conserved seuquences will be verified using EMSA (electrophoretic mobility shift assay ). These results may be contributed in revealing the molecular mechnism of secondary cell wall synthesis regulated by MYB, and provide the base for molecular breeding of trees.
