中文摘要: Piwil2是特异性干细胞表达的特异性蛋白,具有抑制凋亡,影响炎症发生等多种生物学作用。本研究在以往间充质干细胞治疗糖尿病肾病研究基础上,假设通过piwil2定向转染至肾组织,使肾组织具有某些干细胞特性,可能重新具有某些自我修复潜能,从而起到对糖尿病的治疗作用。本项目采用链脲佐菌素干扰制作的糖尿病肾病动物模型,以及人足细胞模型。在细胞层面及动物层面转染Piwli2,采用间接免疫荧光,Westernblotting,CHIP,RT-PCR等分子生物学技术,观察Beclin1、LC3,STAT3,P53、mTOR等蛋白的表达。预期转染piwli2可以通过抑制细胞凋亡,促进细胞自噬发生,改变原有的糖尿病肾病中细胞程序性死亡失衡,而对糖尿病肾病起到治疗作用。

英文摘要: piwil2 is a stem cell-specific expression protein that play a central role in inhibition apoptosis level and development inflammation. Beside on previous study toward treament diabetic nephropathy with stem cell,our hypothesis that renal tissue can get stem cell fuction ,can get an ablility of self-healing,that may paly an improtant role as diabetic nephropathy treament though targeted transfection MRNA of piwil2. With the model of rat induced by streptozotocin and human podocyte, we examined the level of Beclin1,Lc3, STAT3,P53 and mTOR with westernblotting,CHIP ,RT-PCR and so on under targeted trabsfection MRNA of Piwil2.We expected that piwil2 may play an imporant role as diabetic nephropathy though inhibit apoptosis and activition autophagy thus charge the imbalance of programmed cell death.
