中文摘要: 软骨组织不含血管且细胞组成单一,缺乏自我修复能力,损伤后无法修复。鹿茸软骨内含丰富的血管,不仅能够有效的修复损伤,而且能够完全再生。研究显示,鹿茸软骨除含血管外,具备一切软骨组织的特性,为真正的软骨组织。因此,为我们研究软骨内血管生成与损伤修复提供了优秀的模型。本课题前期发现,鹿茸软骨内血管的生成并不是生茸组织(鹿茸软骨的来源)固有的特性,而是由某些外部因子调控的。通过改变生茸组织的外部环境,鹿茸软骨也可以由含血管软骨转变为无血管软骨。为阐明其调控机制,拟利用鹿茸软骨组织的这种生物学特性,建立研究含血管软骨和无血管软骨相互转变的活体模型,通过对两种不同表型软骨的转录组测序和注释及差异基因表达分析,鉴别与鹿茸软骨内血管形成和消失相关的调控因子,并验证相关因子的功能,阐明鹿茸软骨表型转变的调控机制,为临床上开辟一种软骨损伤的内源性修复途径奠定基础。

英文摘要: Cartilage tissue is avascular and consisted of one cell type. Cartilage is inability to repair even minor injury due to its avascular nature. Antler cartilage is unique in that it can effectively repair its damage as it is richly vascularized. Studies have shown that antler vascularized cartilage is true cartilage tissue. It has potential for application as a valuable model for researches on angiogenesis and cartilage repair. Studies have shown that antler stem cells, from which the vascularized cartilage derives, can form avascular cartilage when transplanted subcutaneously into nude mouse or diffusion chamber, suggesting that vascular or avascular state of antler cartilage is dictated by extrinsic factors. But the regulation mechanisms are not clear. In this project we will utilize the antler cartilage development feature to construct an in vivo model, isolate and identify the molecules that regulate this phenotype switch of antler cartilage. It may help us to clarify angiogenesis mechanisms of antler cartilage. Understanding the mechanisms underlying this phenotype switch may help us to devise a way to trigger the intrinsic repair of cartilage, which would have great potential for cartilage repair in the clinic, such as osteoarthritis.
