中文摘要: 通过营养调控预防或缓解免疫应激已成为了现代仔猪培育的重要手段,前期研究表明,姜黄素作为天然活性物具有重要的免疫调控作用,然而在畜禽上的研究仍十分缺乏,其深层次机理尚未清楚。本项目探讨姜黄素对免疫应激仔猪肠道生长发育、结构形态和生理功能的影响;进一步分析姜黄素对肠道黏膜T细胞和树突状细胞的数量、免疫表型、增殖和分化成熟,免疫球蛋白和细胞因子水平,及SOCS、AP-1和TLR等基因和Akt/mTOR信号通路表达的影响;通过体内外试验,以表观遗传组学为体系,利用MeDIP、Chip和高通量测序获取仔猪肠道全基因组DNA甲基化谱、组蛋白修饰谱、miRNA表达谱的数据,深入寻找和分析差异表观修饰位点,整合相关基因、信号通路和表观遗传信息,构建网络图,确定关键基因和表观因子,阐述姜黄素对仔猪肠道免疫功能调控的表观遗传学机理,确定新的营养调控靶点,形成新的学术观点,为仔猪肠道免疫调控新技术提供理论依据。

英文摘要: Nutrition regulation to prevent or relieve the immune stress has become the important means of modern piglets production. Our early studies showed that curcumin as natural compounds plays an important role in regulating immune function, however, little is known about piglets, especially for its mechanism of immune regulation. We will investigate the effects of curcumin on the development, morphology and function of intestine in weaning piglets suffered immune stress. We will further investigate the amount, immunophenotype, proliferation, differentiation and maturation of T cells or dendritic cells, the levels of immunoglobulin and cytokine, and the expression of SOCS, AP-1, TLR and Akt/mTOR signal pathway; Through the experiment in vivo and vitro, using MeDIP, Chip and deep sequencing, genome-wide DNA methylation, Histone modification and microRNA expression profile are determined in intestinal mucosa of weaning piglets. In order to determine the key relative genes and epigenetic factors, the information of DNA methylation, histone modification, microRNA expression profile, SOCS, AP-1, TLR, NF-κB and Akt/mTOR signal pathway will be analysed, and then their network diagram will be built. The results may contributine to the better understanding of the epigenetic mechanisms of curcumin on regulation of intestinal mucosa immunity in weaning piglets, and suggest a potential strategy for enhancing neonatal immune response using curcumin as a new nutritional target.
